Drug intelligence / Profile preview
[161Tb]Tb-J3-7D12-Fc is a novel interleukin-1 receptor accessory protein (IL1RAP)-targeted radioimmunoconjugate being developed for the treatment of Ewing sarcoma (EwS). It consists of the antibody fragment 7D12-Fc, which targets IL1RAP, conjugated via the crown-ether-derived chelator J3 to the beta- and Auger-emitting radioisotope terbium-161 ([161Tb]Tb3+). IL1RAP is highly overexpressed on the surface of Ewing sarcoma cells compared to healthy tissues, making it a promising therapeutic target. The J3 chelator allows for conjugation and radiolabeling under mild, room-temperature conditions, preserving the integrity of the thermolabile antibody fragment. Preclinical studies have demonstrated that [161Tb]Tb-J3-7D12-Fc exhibits high binding affinity, specificity, and significant tumor accumulation in vivo, with ongoing studies evaluating its therapeutic efficacy in tumor-bearing murine models.
